具体而言,该方法通过将目标细胞和药物在不同温度下共培育一定时间后裂解,与药物结合的蛋白会因为热稳定性提高而更不容易发生变性而沉淀,随后通过离心把沉淀的未结合蛋白去除,最后通过蛋白免疫印迹法(Western Blot)或其他蛋白检测技术检测上清液内的蛋白即可获知与该药物结合的蛋白谱,从而研究其药靶相互作用及相应脱靶蛋白。
